I would definitely start at the beginning and go through. I believe you already know my thoughts on Castiel/S4 (or at least I remember bitching in the apartment about them on several occasions), and I think you miss a lot skimming through the first three seasons. (The third is my favorite season btw, but that's a generally unpopular opinion.) I know you're partly (mainly?) getting into SPN because of the theology, but I really think that's the least interesting/least coherent part of SPN. And I say this as someone seriously considering Religious Studies as a major.
Then again, I suppose the argument could be made that the theology is interesting because it is so poorly fleshed out. *g*
